A Mechanism Study of NMDAR1 in A Rat Alzheimer Disease (AD) Model BABT

ABSTRACT This study aimed to investigate the expression and mechanism of N- methyl -D- aspartate receptor 1 (NMDAR1) in the pathogenesis of Alzheimer disease (AD). Eighty adult Wistar rats were randomly divided into 4 groups (n=20 each) to receive an injection of 0, 5, 7 and 10 μl of 1 μg/μl amyloid-β 42 (Aβ1-42) in the hippocampus. Twenty rats in normal control group were injected with equal volume of saline. After 10 days, the hippocampus was isolated from 5 randomly selected rats in each group. The NMDAR1 protein and mRNA expression was determined by immunohistochemical staining and qRT-PCR. The aquaporin-1 (AQP-1) mRNA expression was also measured by qRT-PCR. A method to improve leaf succulence quantification BABT

Leaf succulence has important physiological and ecological implications. Currently it is quantified by Delf's index (fresh weight/leaf area) and fresh weight/ dry weight ratio. Both indeces are reconsidered and a new index is proposed. Shade and sun leaves from terrestrial, hemiepiphytic and epiphytic aroids were studied. Delf's formula, which does not consider dry weight, overestimated leaf succulence. As fresh weight / dry weight ratio (fw / dw) does not consider leaf area, plants with the same fw / dw ratio were more than five times different in area. A methodological proposal for quantifying environmental compensation through the spatial analysis of vulnerability indicators BABT

The aim of this work was to propose a methodology for quantifying the environmental compensation through the spatial analysis of vulnerability indicators. A case study was applied for the analysis of sand extraction enterprises, in the region of Descalvado and Analândia, inland of São Paulo State, Brazil. Environmental vulnerability scores were attributed for the indicators related to erosion, hydrological resources and biodiversity loss. This methodological proposal allowed analyzing the local alternatives of certain enterprise with the objective of reducing impacts and at the same time reducing the costs of environmental compensation. A model describing the relationship between electrical conductivity and forage dry matter content BABT

The correlation between dry matter (DM) content, electrical conductivity (EC) and ash content was found to be linear in grass and alfalfa during drying. While minerals in grass and alfalfa were very stable components during drying, their concentrations and EC increase accordingly; therefore, the EC value could serve as a measurement to determine DM content. Results indicated that EC measurement in grass and alfalfa during drying was simple, accurate, fast and easily repeatable, and could serve as an alternative method for DM determination.

A model for the study of skeletal anomalies in rat fetuses BABT

The aim of this study was to validate a model of skeletal anomalies in rat fetuses by the administration of ketoconazole (80 mg/kg) to pregnant rats during organogenesis. Bones of the head, trunk and anterior and posterior limbs were examined for detection of anomalies. Statistical differences regarding the number of fetuses and postimplantation resorptions, and fetal and placental weight were significant. The frequency of skeletal anomalies in the head, trunk, and anterior and posterior limbs in the ketoconazole-treated group were also significant when compared to the control group. A modified method for the turbidimetric assay of nisin BABT

Nisin is an antibacterial peptide produced by Lactococcus lactis subsp. lactis that shows a broad spectrum of inhibitory activity against gram-positive microorganism including bacterial spores. Methods for nisin activity estimation in solution is redefined. In this work a new turbidimetric assay method has been described making possible to assay nisin satisfactorily in total incubation period of approximately six hours.

A new alternative to produce gibberellic acid by solid state fermentation BABT

Gibberellic acid (GA3) is an important hormone, which controls plant's growth and development. Solid State Fermentation (SSF) allows the use of agro-industrial residues reducing the production costs. The screening of strains (four of Gibberella fujikuoroi and one of Fusarium moniliforme) and substrates (citric pulp, soy bran, sugarcane bagasse, soy husk, cassava bagasse and coffee husk) and inoculum preparation study were conducted in order to evaluate the best conditions to produce GA3 by SSF. A new biosurfactant produced by Candida glabrata UCP 1002: characteristics of stability and application in oil recovery BABT

The production of a new biosurfactant by Candida glabrata UCP1002 was studied to evaluate the influence of the concentration of the cotton seed oil, glucose and yeast extract. The dynamics of the growth and surfactant production were showed for all the cultivation conditions studied. The best emulsification of the n-hexadecane, quantified by the emulsifying index was observed in the medium containing 7.5% cotton seed oil, 5% glucose and 0.3% yeast extract. The isolated biosurfactant showed a CMC of 2.5% and the surface tension at that point showed to be 31mN/m. A new enzymatic process for the treatment of phenolic pollutants BABT

This work aimed to develop a new economic enzymatic process to treat the phenolic pollutants using pure tyrosinase in stirred vessel and adopting temperature (T), pH, rotational speed (N), initial phenol (C P,o) and enzyme (C T) concentrations as independent variables. Experimental data of the residual phenol concentration (C P) were used to calculate the oxidation efficiency (η), initial oxidation rate (-r o) and time required to reach the end of reaction (t) that were selected as the responses. Under the optimal conditions (T = 45°C, pH 6.6, N = 400 rpm, C P,o = 100 ppm and C T = 50 U/mL), η was 88.1%, -r o = 10.2 mg L-1 min-1, t = 40 min. These results suggested that tyrosinase-rich crude extracts from vegetable byproducts could be quite promising.

A new mini box corer for sampling muddy bottoms in antarctic shallow waters BABT

A new Mini Box Corer (MBC-GEAMB) was developed for bottom sampling in Antarctic shallow waters down to 100 m depth from a small vessel. It consists of a detachable stainless steel box with a total sampling area of 0.0625 m², and a closing arm with a detachable blade without an external frame. MBC allowed stratified bottom sampling and good quality samples comparable to those obtained through diving. A comparison between the MBC-GEAMB and a 0.056 m²van Veen grab (VV) was undertaken for the benthic macrofaunal composition in Admiralty Bay, King George Island (Antarctica). A nonspecific primer anchored PCR technique for chromosome walking BABT

A chromosome walking method was improved in this work. The new method was named as nonspecific primer anchored PCR (NPA-PCR). Nested gene specific primers were designed based on the known region and long random primer using degeneracy oligonucleotides for nonspecific anchoring. Annealing temperatures were varied to control the priming. Target sequences were obtained by PCR with random primer and gene-specific primer. Nonspecific sequence with long random primers at both ends formed stem loop structure due to inverted terminal repeats. A Novel Raw Starch Hydrolyzing Thermostable α-Amylase Produced by Newly Isolated Bacillus mojavensis SO-10: Purification, Characterization and Usage in Starch Industries BABT

ABSTRACT The aim of this study is the production, purification, and characterisation of thermostable raw starch hydrolyzing α-amylase produced by Bacillus mojavensis SO-10. The maximum production conditions of α-amylase were found at 36th hour, 35 °C and pH 7.0. We utilized three steps to purify the thermostable α-amylase and as a result, 34-fold and 18% yield were obtained. The molecular weight of purified α-amylase was determined as 73 kD. The Km and Vmax rates were detected as 0.010 mM and 3.38 µmol min−1, respectively. This purified α-amylase exhibited the highest activity at pH 5.0-6.0 and 70 ºC and showed stability over a wide variety of pH and temperature at 4.0-8.0, and 40-50 ºC, respectively. A practical culture technique for enhanced production of manganese peroxidase by Anthracophyllum discolor Sp4 BABT

In this study, different growth conditions of Anthracophyllum discolor Sp4 including the effect of agitation, additions of lignocellulosic support, inducer and surfactant were evaluated on the MnP production in Kirk medium using a culture system made up of the tubes containing the glass bead . The highest MnP production (1,354 U/L on day 13) was obtained when the medium was supplemented with wheat grain and 0.25 mM MnSO4 as inducer, under static conditions at 30°C. Two isoenzymes were purified (35 and 38 kDa respectively). MnP presented a maximal activity in the pH range between 4.5 and 5.5, a relatively high temperature tolerance (50ºC) and a high catalytic activity for 2,6-dimethoxyphenol and hydrogen peroxide.

O autor relata trabalhos experimentais que o levaram a concluir pela existência da galactoquinase em tecidos animais, explicando desta maneira, que a primeira fase do metabolismo intermediário da galactose nos mesmos é a sua esterificação, em presença do ATP e da enzima em questão, ativada pelo magnésio, para galactose-1-fosfato.

A rapid and reliable method for the clonal isolation of Acanthamoeba from environmental samples BABT

Acanthamoeba are abundant in a wide range of environments, and some species are responsible for cutaneous infections, keratitis, and granulomatous amoebic encephalitis (GAE). The conventional detection and isolation of amoeba from clinical and environmental samples involves sampling and culture on non-nutrient Ágar medium. Although efficient, this system requires several transfers in order to eliminate contaminants, and is not appropriate for the isolation of individual amoeba from samples with a biodiverse community. In this study we propose an alternative method for the isolation of monocystic clones of Acanthamoeba. The propose method involves sampling, enrichment, encystment induction, and direct cysts micromanipulation and culture on Ágar plates.

A Reassessment of the in vitro total protein content determination (TPC) with SIRC and 3T3 cells for the evaluation of the ocular irritation potential of shampoos: comparison with the in vivo Draize rabbit test BABT

The aim of this work was to determine the correlation between results obtained from the Draize test and from the Total Protein Content Determination (TPC) to assess the ocular irritancy potential of 20 shampoos. For TPC, two established cell lines (SIRC and 3T3) were used. The concentration that induced 50% inhibition relative to controls (IC50) was calculated for each product. Among shampoos tested with SIRC, only one had a false positive result. However, for the 3T3, three false-negative results were found. Pearson coefficient related to the in vivo value of maximum average score (MAS) was -0.58 (p=0.007) with SIRC and -0.73 (p=0.007) with 3T3. ABSTRACT Ferulic acid (FA) is a phenolic compound with well-known antioxidant potential that can be used as a promising anti-inflammatory and anti-cancer molecule. Furthermore, it has been reported to have neuroprotective activity. One of the main problems, which limit its clinical use, is its low bioavailability when administered orally. This limitation can be circumvented by changes in their structure and/or for preparing lipid-based formulations.
